What is Retatrutide and why is it significant in metabolic research?
Retatrutide is a novel synthetic peptide developed by Eli Lilly as the first investigational triple agonist of the glucagon-like peptide-1 (GLP-1), glucose-dependent insulinotropic polypeptide (GIP) and glucagon receptors. Unlike single-agonist incretin mimetics such as semaglutide or dual agonists such as tirzepatide, retatrutide engages all three metabolic receptors in parallel — producing a markedly broader signalling profile in in-vitro models of energy expenditure, adipogenesis and hepatic lipid metabolism. Phase 2 data published in the New England Journal of Medicine (Jastreboff et al., 2023) reported a mean 17.5% body weight reduction at 24 weeks at the highest dose cohort, positioning retatrutide as the most potent investigational compound in its class. Phase 3 trials commenced in 2023.
