About L-Carnitine
L-Carnitine is a small quaternary ammonium compound, C7H15NO3, 161.2 Da, not a peptide. It is the R-enantiomer of carnitine, made in the body from lysine and methionine, and is studied for its role in moving long-chain fatty acids into mitochondria. L-Carnitine in the research literature
The function of carnitine stayed open until Fritz and colleagues, 1959, showed in Science that it governs long-chain fatty acid oxidation in muscle preparations. The Bremer 1983 review in Physiological Reviews set out the carnitine shuttle as it is still taught: carnitine palmitoyltransferase I on the outer mitochondrial membrane, the carnitine-acylcarnitine translocase, and carnitine palmitoyltransferase II on the inner membrane. Rebouche and colleagues, 2004, cover the kinetics and regulation of L-carnitine and acetyl-L-carnitine metabolism.
More recent work is less tidy. Koeth and colleagues, 2013, in Nature Medicine, traced gut microbial conversion of L-carnitine to trimethylamine N-oxide, and that paper is the reason carnitine now appears in microbiome and TMAO studies. Hathcock and colleagues, 2006, is the formal risk assessment for carnitine as a nutrient. Stereochemistry matters throughout: assays use the R-enantiomer, levocarnitine, because the D form is not handled by the acyltransferases in the same way.
